FUNCTION OF ERYTHROCYTIC TRANSPORT
OF GLUCOSE
Erythrocytes which are the most
numerous part of regular elements of blood have disk-shaped, concave-concave
form. Hemoglobin composes the basis of contained erythrocyte. Basic function of
erythrocytes not only supply tissues with oxygen and other substances but also
removal from them carbonic acid.
According to all available information
transport function of erythrocytes is not limited on the surface
membranes. Erythrocytes adsorb from
plasma glucose, amino acids, proteins, lipids and transfer them to the tissues.
Glucose is basic and most important
energy material for maintenance of metabolic processes in the organism of a
human and animals. Especially intensively it is consumed by tissues of central
nervous system, muscular system etc. More
than a half of the energy which expends our organism is formed due to the
oxidation.
Consequently, glucose is the most valuable nutrient for the majority
of cells which concentration plays
leading part in metabolism and determines by
balance between its expense and arrival from the food or as a result synthesis
in the organism. Maintenance
of glucose at the proper level is essential and determining for the
vitality.
Disadvantageous
action of environment and deficient supply of tissues by oxygen the organism of
a human and animals, lead to hypoxia.
Continuous
energy consumption is the necessary condition of the vital activity of any
biological structure. Hypoxia not only
complicates process of disease connected with functional insufficiency of
respiratory and cardiovascular systems, diseases of liver, kidneys, endocrine
system, etc, but also is defines its outcome.
The manifestations of hypoxia depend on etiology, degrees, speed of
development and duration of hypoxic state, reactivity of the organism, in this
case occur reduction in the delivery of oxygen to tissues, damage of metabolism
and structure of cells, and also adsorptive- transport properties of
erythrocytes [1, 2, 3, 4, 5, 6, 7, 8, 9].
The purpose of the present
investigation is study of erythrocytic transport of glucose with hypoxia in experiment.
Materials and methods
Experiments
were conducted on 64 pedigreeless adult dogs weighing 12-18 kg, which were
divided into 2 groups: the 1st group – control group, and for animals of the
2nd group was invented the model of hypoxia.
In experiment blood was separated
into plasma and erythrocyte sediment to determine the concentration and content
of glucose in blood plasma and on surface of erythrocytes.
Blood was centrifuged for 5 minutes
at 1500 rpm for adsorbed substances from the erythrocyte sediment. Thus one
part of erythrocytic mass diluted in a volume ratio to three parts of 3%
solution of sodium chloride.
The obtained single washing from
erythrocytes (suspension) gently mixed and incubated at 37 °C for 5 minutes for
the full flush of molecules associated with membrane of erythrocytes. Then
erythrocytic suspension was centrifuged at 1500 rpm for 5
minutes. As a result of this procedure in erythrocyte washout passed the
substances adsorbed on membrane of red blood cells and supernatant (washing)
was transferred into clean tubes [of 10, 11].
Biochemical blood analysis was conducted on the unit “Automatic
biochemical analyzer “BioSystems A-25” (Spain)”.
Static processing has been done on the computer. The criteria of
Styudent t - was significant at P <0,05.
Results of study and discussion
As a result of studies in the 2nd
experimental group of animals on a background of hypoxia, where the number of
erythrocytes significantly increased at 18,4%,
and hemoglobin level of 9,23% as compared with the control group (Pic.
No. 1 and no.2).

According
to study in blood plasma in the 2nd group in comparison with the control group
observed a significant increase of glucose to 6,79+0,09 mmole/liter (control
5,65+0,10 mmole/liter). The percentage of glucose content of glucose in the
washings significantly increased by 39,6% compared with the control group.
Sample
of rates and adsorptive-transport function of erythrocyte on background of
hypoxia, adsorption and transfer of substances by red cells of the blood is the
mechanism in response to exposure of unusual factors of environment to organism.
Results
of researches indicate that during experimental hypoxia in plasma increases not
only red blood but also adsorptive-transport function of erythrocytes.
