The Activity Increase of the Peritoneal Macrophages of
the Mice, infected with M.tuberculosis H37Rv,
treated with the preparation “GLYCYRFIT”
from Glycyrrhiza
glabra.

The studies showed, that per os introduction of the
preparation GLYCYRFIT from Glycyrrhiza glabra root to the mice with
intraperitoneal M.tuberculosis-H37Rv infection significantly
increases the functional activity of peritoneal macrophages in absorptive
capacity and the activity increasing of one of the main microbicidal functions
of phagocytes – myeloperoxidase system.
Such characteristics of
the fito-preparation allow us to recommend using it in the treatment of
tuberculosis.

Исследования
показали, что введение препарата «ГЛИЦИРФИТ» из корня Glycyrrhiza glabra per os мышам, зараженным
внутрибрюшинно Mycobacterium tuberculosis H37Rv, повышает
функциональную активность перитонеальных макрофагов по показателям
поглотительной способности и повышению активности основной бактерицидной
функции фагоцитов – миелопероксидазной системы. Такие свойства фитопрепарата позволяет рекомендовать его использование
в комплексной терапии туберкулеза.

Introduction
The
currently used anti-TB drugs with etiotropic and antibiotic effect have been
applied for a long time because of their effect
on the etiological agent - Mycobacterium tuberculosis. They can cause some side
effects, such as disruption of metabolic processes and functional status of
some organs. The patients with pulmonary
tuberculosis have defect in
the functional activity of peripheral blood monocytes and the oppression of their absorptive capacity.

Herbal preparations have a number of positive properties: a high
efficiency with low toxicity, a complex harmonizing effect on an organism, a
broad spectrum of effects and the relative cheapness.
Objectives
The main objectives of this study are: the evaluation of
the effects of GLYCYRFIT preparation from the root of Glycyrrhiza glabra on the functional activity of peritoneal macrophages
(PM) of the stimulated mice with intraperitoneal M.tuberculosis-H37Rv
infection; the evaluation of the
possibility of GLYCYRFIT application to enhance the functional activity of peritoneal macrophages during the infection
with M.tuberculosis Н37Rv.
Materials and methods
The object of the
research was the preparation GLYCYRFIT from Glycyrrhiza
glabra; CBA mice with intraperitoneal M.tuberculosis-H37Rv
infection.
During the experiment we used 100
CBA mice, synchronized by weight and
kept under standard vivarium conditions. All the animals were divided into 2 groups –
treated and not treated with the preparation GLICIRFIT from Glycyrrhiza
glabra. The peritoneal macrophages were
extracted according to the standard method. The mice of all groups were initially
stimulated by intraperitoneal injection of 2% solution
of peptone-casein (Serva) at 5 ml per a mouse, then the mice were injected
intraperitoneally with 0.6% solution
of hydrogen peroxide - 2 ml per
a mouse – in order to reduce the bactericidal activity of basic
myeloperoxidase phagocyte system. After that both groups of
mice were injected intraperitoneally
with suspension of the laboratory strains culture of M.tuberculosis-N37Rv,
diluted to a final concentration of 106 microbial bodies per
1 ml. Daily, for
7 days the first group of the mice were
injected through a catheter into
the esophagus with 0.2 ml/kg GLYCYRFIT preparation from root of Glycyrrhiza glabra. Functional activity of peritoneal macrophages
(PM) was evaluated at the 1st,
2nd, 3rd, 4th,
5th, 6th, 7th days, using the test for the definition of absorptive capacity and the level
of myeloperoxidase activity. For the determining the absorptive capacity
of peritoneal macrophages the phagocytic index (PI) – percentage of phagocyte
peritoneal macrophages to the total number of
cells and phagocytic number (PN) – the average number of mycobacterium, absorbed by a single cell, was taken into account. The
activity of myeloperoxidase in
peritoneal macrophages was determined through semi quantitative method.
Results and discussion
The
study of the functional activity index of peritoneal macrophages (PM) in first
group mice showed a significant increase of phagocytic index (PI) and
phagocytic number (PN) after daily injection of the preparation GLYCYRFIT from
Glycyrrhiza glabra in comparison with
the mice, not treated with "GLYCYRFIT".
This is due to the fact that peritoneal
macrophages (PM), being active phagocytic cells, reduce the number of
M.tuberculosis H37 Rv, which were injected to animals at the beginning of the
experiment. At the same time, the increase of
myeloperoxidase (MP) activity was found out in peritoneal macrophages
(PM) of the group of the mice, treated with "GLYCYRFIT".
In this case, the indicators of myeloperoxidase (MP) activity on the 7-th day
dosing was significantly higher than the same index in intact mice (p
<0.05). The indexes of myeloperoxidase (MP) activity in peritoneal
macrophages (PM) of the intact mice were low without preparation treatment;
during the absorption and the possible persistence of M.tuberculosis H37Rv,
were going down to the 7th day.
Thus, there is clearly a significant increase of
the phagocytic index (PI) and the phagocyte number (PN), as well as
myeloperoksidase (MP) activity in groups of mice, treated with
"GLYCYRFIT" [1, 4, 5, 6].
The activity of immunological mechanisms in the
mice organisms infected with M.tuberculosis H37Rv significantly
increases to the 7th day after the regular feeding with
"GLYCYRFIT" from the root of Glycyrrhiza glabra. It is observed more
rapid recovery of this group of animals to the comparison with the animals,
treated with traditional anti- M.tuberculosis antibiotics.
Previously, we showed in vivo on Shepard leprosy
model an active antimicrobial effect of horseradish peroxidase on mycobacterium
leprous [4]. An extract of licorice
root (Glycyrrhiza glabra) provided similar effects on M.leprae; as well, it has
a bactericidal action in vitro on M.tuberculosis H37Rv, namely it
possesses an expressed antitubercular activity.
Conclusion
Thus, the preparation "GLYCYRFIT" from
the root of Glycyrrhiza glabra significantly increases the functional activity
of peritoneal macrophages (PM) according to indicators of absorptive capacity
and to the activity increase of myeloperoxidase (MP), as a part of one of the
main microbicidal systems of phagocytes - myeloperoksidase system.
Some plant substances of licorice root
Glycyrrhiza glabra, including antioxidant components bioflavonoids, besides
stimulation of endogenous cortisol and
antimicrobial activity possess the positive impact on the humoral immune
system.
The positive characteristics possessed by
phytopreparation, including "GLYCYRFIT" from Glycyrrhiza glabra,
allow us to recommend the using this phytopreparation in the treatment of
tuberculosis[1, 7].
