THE MASS-EXCHANGE
AND AERATION IN BIOREAKTOR'S
The
utility model belongs to biotechnology and may be used in the microbiological,
pharmaceutical and food industries for cultivation of cells, probiotic
cultures, the culture of lactobacillus, and for needs of medicine and clinical
researches.
The
known plant for cultivation of microorganisms, which consists of two chambers,
which are connected in the lower part by flexible piping. These chambers have
air filters and reversible drive for their reverse-forward movement in the
vertical plane (À.c. The USSR ¹1131899, Ñ12 Ì1/00, 1984).
The
weakness of this plant are the inadmissible big dimensions, which are causing
by the necessity of movement of the chambers in the vertical plane.
Also
there is known the apparatus for cultivation of cells (AC), which contains a
body with the technological union couplings and the hollow shaft, which is
placed on the axes of the body of impulsive drive with a blushing, added with
the mixing element in form of four-line hinge with shoulder-blades on ends, the
filtering element connected with the interstice of the shaft and mobile hinge
bush, and also an aerator (A.c. The USSR 1633814, C12 M3/00, 1995).
This AC
is the closest to the utility model for it’s technical essence and achivable
effect. The weakness of it is the deffisient productivity of making of the
product.
The
noted weakness is explaining by condition, when the diminishing of number of
turns of the shaft, the intensity of mixing and cells of biomass goes down and
do not provide in largo manum by oxygen which slows their development. The
opposite, with the increasing of number of turns of the shaft, the mixing
element inflicts physical harm to microorganisms, limiting the rates of their
development which also interferes with growth of the productivity. Except this,
a vehicle has an elaborate design which can be considered as another his
defect.
The
basis of utility model is the task to improve the vehicle for the cultivation
of cells by modification of form and structure of the motion of mixing element,
and as a result the necessary degree of active mass exchange is providing and
diminishing of degree of risk of damage of the shells of microorganisms.
Providing of these terms serves to increase the productivity with simultaneous
simplification of construction.
The
given task is solving by that in ÀC which contains the cylindrical case with
the technological branch pipes, a shaft with the plug placed along an axis of
the case which is attached with a
mixing element, an aerator, and also a reversive drive, according to utility model
there is a new fact of presence of contacting surfaces of a shaft among
themselves and the plug in the form of carving connection, and a mixing element
- in the form of a disk which is saved from the reference around the vertical
guide.
The outlined
difference allows to intensify process of hashing of a biomass without damage
to the cells which raises productivity, i.e. increases quantity of an exit of
probiotics which structure includes live cells of producers. Simultaneously
with it the design as the mixing element has simple and reliable in work the
geometrical form becomes simpler.
On fig.
1 the offered device for the cultivation of cells is represented schematically.
AC contains the
cylindrical case 1 with a branch pipe 2 for introduction of a nutritious liquid
and a sowing material, a branch pipe 3 with àthe aerator 4, a branch pipe 5 for
removal of the nutrient liquids, and a branch pipe 6 for removal of the
fulfilled gas. Along a axis of the case 1 the shaft 9 with the plug 10 is
located and is attached to a motor-reducer 7 with the command reversing device
8. On this plug 10 a mixing element –the disk 11 in the form of established
with a clearance "δ" concerning the walls of the case 1. The
contacting among themselves surfaces of a shaft 9 and plugs 10 are executed in
the form of carving connection (screw pair), and the disk 11 counts fixed in
the case 1 vertical guide 12.
The device works in
the following way. In preliminary sterilized AC to the case 1 enter through a
branch pipe 2 nutrient liquid and a sowing material (subculture) then in the
aerator 4 gas for aeration of cultural medium arrives. The command device 8
engages, which signal puts in action the motor-reducer 7 and a shaft 9 which
within 8 courses set by the command device "Í" gives back and forth
motion to the plug 10 and attached to it and saved from the reference to a disk
11guide 12.
Moving along a
shaft, the disk 11 would carry out the mass exchange of spilling over biomass
through a backlash without the damage to the cells owing to what the conditions
for more intensive growth of productivity of process are created.
The
absence in the offered construction also assists to reduction of the risk of
damage to the cells, and also to a design of rotating movement of a disk 11
that takes place in a prototype, and its simple form leads to design
simplification as a whole. Upon termination of cultivation process, the
motor-reducer 7 is stopped, and a finished product merge through a branch pipe
5 for further using.
