Assessment of adaptation with using apoptotic markers CD95 and APO2.7
Adapting
to the resort - the process of adaptation of the organism to new environmental
conditions in which there is treatment and rest. There
are characteristic adaptation processes in children,
prolonged and frequent episodes of arriving at the spa treatment at health
resorts of Crimea from various regions of Ukraine.
It is known that the degree of stress on physiological
systems due to the contrast between the climatic conditions of place of
residence. At a certain category of children is seen dramatic disruption of
adaptation, the emergence of stress that can affect the immune system. The aim
of our study was to determine the immune status of children with the syndrome
dysadaptation, as well as monitoring the status of cellular immunity using
apoptosis markers CD95 and APO2.7.
We
examined 25 children of both sexes
aged from 8 to 16 years old, located on the sanatorium stage of
rehabilitation in sanatoria "Jubilee" and "Emerald" Evpatoria and classified as long and often
ailing, mainly chronic naso-pharyngeal region.
Adaptation
assessment in children categories of long and frequent episodes included the
analysis of peripheral blood of patients with the use of the following markers:
CD95 - cell-surface receptor that can trigger apoptosis in it after its
interaction with Fas-ligand or monoclonal antibodies to Fas, APO2.7 - for
determine protsentsnoy of cells that have undergone apoptosis. Method of
assessing the state of adaptation using markers CD95 and APO2.7 includes the
use of flow cytometry and marker-receptor CD-95 as indktora apoptosis, and
additionally used a marker antigen APO2.7 (also known as 7A6), which appears on
the membrane of mitochondria in the early stages of apoptosis. Expression of
APO2.7 detected in the early stages of apoptosis. APO2.7 also can be detected
in cells after exposure to radiation, drugs or binding of CD-95, Fas-ligand.
Living cells are negative or slabopozitivny to this antigen.
The
analysing of
the data indicates that the children at early stage of deviation from the normal processes of
adaptation have increased apoptosis (increase in CD95 expression and APO2.7,
indicating that the development processes dysadaptation). The degree of
degradation of cellular immunity is reflected in an inverse correlation between
indicators of cellular immunity (reduction of CD3 + and CD4 +) and indicators
of apoptosis (increased CD95 and APO2.7).
The analysis of the data that children with abnormal states
of adaptation by identifying markers of apoptosis
of peripheral blood neutrophils has been a strengthening of
apoptosis. Investigating the correlation between the level of cellular
immunity, indicators of apoptosis, we noticed that there is a close inverse
correlation (r> -0,5) between the indices of cellular immunity (reduction of
CD3 +, CD4 +) and indicators of apoptosis (increase CD95 and APO 2.7 ).
Thus,
the deviation from the norm of the state of adaptation by measuring apoptosis
of peripheral blood neutrophils was in the clear depending on the treatment
measures at the stage of sanatorium rehabilitation.
